Does the load on a tire affect the pressure reading on a tire? I know for a fact my truck weighs about 1500 pounds more than it did when it left the factory (utility bed loaded with stuff and tools). Am I getting an accurate on my tires? If this is the case how can a tire company put a rating on a tire? Should I jack up my truck to check the tires?

Nope, no problem.

40 psi is 40 psi, wherever it is or whatever it's doing, whatever it's holding up.

Ditto. The only way to change the pressure in the tire is to somehow change the volume (overall size) of the tire, which you can't magically do. When the tire pressure is low you get the the sidewall "bulge" which is keeping the overall volume the same as if it was at max pressure and no bulge (straight sidewalls).
